PURPOSE:Crosslinked hyaluronic acid that is obtained by crosslinking hyaluronic acid (abbreviated to HA) or its salt with a polyfunctional epoxide, thus showing high stability to enzymatic hydrolysis and nonenzymatic oxidative and reductive decomposition. CONSTITUTION:HA or its salt is dissolved in an aqueous alkali (such as NaOH of 12-14pH) so that its concentration usually becomes over 1.0%, then, a water- soluble organic solvent such as methanol or acetone is added until the amount exceeds 50% of the total solution volume. Subsequently, a polyfunctional epoxide such as epichlorohydrin, or 1,2-bis(2,3-epoxy)ethane is used to effect crosslinking so that the number of crosslinkings exceeds 5 per 1,000 disaccharide units of glucuronic acid and N-acetylglucosamine to give the objective crosslinked HA. USE:s-Crosslinked HA has analgesic action on arthritis and can pass smoothly through syringe needles despite its high viscosity. is-Crosslinked HA is suitable for use as a support in affinity chromatography.
