<p>PROBLEM TO BE SOLVED: To provide the subject specific support capable of synthesizing various oligonucleotides through maintaining mild acidic or basic reaction conditions and ensuring them to be efficiently and quantitatively released and detected. SOLUTION: This support for oligonucleotide synthesis has a structure of the formula (S is a solid polymeric support; B is a purine base, pyrimidine base or a derivative thereof; a wavy line denotes a spacer with 1-50 atoms bound to the polymeric support without cleavage under acidic or basic deprotection conditions in oligonucleotide synthesis; R8 is a protective group for oxidizable substituent; R7 is an OH-protective group; R9, is H, an alkyl, alkoxy or aryl). An oligonucleotide having a protective group unstable to base is synthesized on the O-atom of the group OR7 of this support followed by deprotection of the protective group with a base and then hybridizing the resultant oligonucleotide with the oligonucleotide complementary to the former to form a double strand and also detect both the oligonucleotides.</p> |
