A method for patterning a substrate used for cell culturing is provided to pattern the surface of the substrate through more convenient and simpler process. And a patterned substrate prepared by the same method and a method for culturing cells on the patterned substrate are provided. The method comprises the steps of: (a) attaching a cover to a partial area of the solid support surface; (b) coating a hydrogel copolymer including repeating units represented by the formulae(1) and (2) on the solid support surface; and (c) removing the cover from the solid support surface. In the formulae, X1 is a single bond, a substituted or unsubstituted C1-10 alkylene, a substituted or unsubstituted C2-10 alkenylene, or a substituted or unsubstituted C6-20 arylene, each R1, R2 and R3 is independently H, a halogen, a substituted or unsubstituted C1-10 alkyl, or a substituted or unsubstituted C1-10 alkenyl, b is an integer from 10-100,000, X3 is a single bond, a substituted or unsubstituted C1-10 alkylene, a substituted or unsubstituted C2-10 alkenylene, or a substituted or unsubstituted C6-20 arylene, R5 is a functional group represented by the structural formula(2-a), (2-b) or (2-d), and d is an integer from 10-100,000.
