Prodn. of viral membrane protein domains

Prodn. of viral membrane protein domains (VMPD) in native form by (i) insertion of a nucleotide sequence for a recognition sequence, e.g. proteases or glycolipid anchors, into an appropriate site of the encoding gene (ii) expression of (i) in eukaryotic cells (iii) subjecting (ii) pref. to a cleavage by a protease which specifically recognises the inserted sequence and cleaves it, and (iv) isolating the desired protein domain from the mixture. Pref. the VMPD are ecto-domains of the HIV env-glycoprotein.The recognition sequence pref. is -(G-P-X)n with n more than 1, pref. 3 and X any aminoacid and the cleavage enzyme pref. is a collagenase.Also claimed are the domains of viral membrane proteins isolated by the process above which are present in native, pref. in oligomer and glycosylated form.
