ANALYSIS OF MONOSACCHARIDE IN SACCHARIDE COMPOSITION

PROBLEM TO BE SOLVED: To provide a method for determining the quantity of monosaccharides such as a sialic acid constituting a saccharide composition. SOLUTION: When simultaneously performing the quantitative analysis of monosaccharides constituting a saccharide composition, this method features the following steps: (1) isolating a sialic acid from the saccharide composition by a sialidase or an acid; (2) transforming the isolated sialic acid into an N- acylmannosamine by a sialic acid aldolase and; (3) hydrolyzing the N- acylmannosamine and saccharide moiety by an acid. A sample can be pretreated in a single container and a ssaccharide composition including a sialic acid can be learned by the analysis such as HPLC of the once.
