摘要 PROBLEM TO BE SOLVED: To provide a microscope system in which a confocal observation using a scanning type laser microscope and a fluorescent observation using evanescent illumination can be simultaneously or switchably performed, and especially even when a focal position in an optical direction on the scanning type laser microscope side is positively changed, the usability for the fluorescent observation using the evanescent illumination is excellent. SOLUTION: A laser beam from an Ar laser 2 is converged onto a sample 25 through a scanning unit 10 and an objective lens 24 so as to be two-dimensionally scanned, evanescent illumination is generated on the sample 25 by a laser beam from a green helium neon laser 3 and a wave front conversion element 14 is arranged on the optical path of the laser beam from the Ar laser 2. Then mechanical positional relation between the objective lens 24 and the sample 25 is fixed on the sample 25 so that a condition for performing evanescent illumination is satisfied and the converged position of the laser beam on the surface of the sample 25 can be controlled by the wave front conversion element 14.
